Source DNA sequence encoding the Human (HLA-A*03:01) AA25-305 [accession# P04439] and the B2M protein AA2-119 [accession# P61769], fused to the mutant G12V KRAS epitope VVVGAVGVGK .This complex includes a C terminal polyHis AVI tag and was expressed in HEK cells, The teramer conformation was achieved through biotinylation of the monomer and then covalently binding the monomers via an avidin biotin reaction.
Molecular weight Recombinant HLA-A/KRAS G12V has a calculated mass of approximately 250kDa. Due to glycosylation migrates as an approximately 260 kDa protein under reducing conditions in SDS-PAGE.
Purity purity was determined to be >95% by nonreducing PAGE and HPLC (see below methods)
Activity The activity was tested in an ELISA assay. Human KRAS G12V (HLA-A*03:01) Tetramer binds Anti KRSA G12V antibody with the EC50 of 0.6μg/ml .
Endotoxin Endotoxin content was assayed using a LAL gel clot method. Endotoxin level was found to be less than 0.1 ng/µg(1EU/µg).
Presentation Recombinant HLA-A*03:01 KRAS G12V complex was lyophilized from 0.2 µm filtered PBS solution pH7.4
Reconstitution A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.
Storage The lyophilized protein is stable for at least 2 years from date of receipt at -20° C.
