DNA sequence encoding the Human (HLA-A*02:01) AA25-305 [accession# P04439] and the B2M protein AA2-119 [accession# P61769], fused to the GP100 (PMEL) peptide YLEPGPVTA .This complex includes a C terminal polyHis AVI tag and was expressed in HEK cells, The tetramer conformation was achieved through biotinylation of the monomer and then covalently binding the monomers via an avidin biotin reaction.
Molecular weight
Recombinant HLA-A/GP100 Tetramer has a calculated mass of approximately 50kDa. Due to glycosylation migrates as an approximately 60 kDa protein under reducing conditions in SDS-PAGE.
Purity
purity was determined to be >95% by nonreducing PAGE and HPLC (see below methods)
Endotoxin
Endotoxin content was assayed using a LAL gel clot method. Endotoxin level was found to be less than 0.1 ng/µg(1EU/µg).
Presentation
Recombinant HLA-A*02:01 GP100 Tetramer was lyophilized from 0.2 µm filtered PBS solution pH7.4
Reconstitution
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.
Storage
The lyophilized protein is stable for at least 2 years from date of receipt at -20° C.
