Source DNA sequence encoding the COVID-19 Spike S protein extracellular domain,[16-1188]R683A (accession # QHD43416.1) fused to a a T4 Folsom trimerzation domain and a C-terminal His AVI tag was expressed in HEK Cells. The furin cleavage site has been mutated at position 683 and 685 so that the protein is not cleaved into the S1 and S2 primary domains by cellular proteases.
Biotinylation The S trimer is biotinylated in vitro at a single site using the AVItagTM biotinylation technology.
Molecular weight Recombinant 2019-nCoV Spike S trimer protein is a protein consisting of the S1+S2 subunits amino acid residues. The protein has a theoretical MW of 140kDa,however due to glycosylation migrates as an approximately 200 kDa protein on reduced SDS-PAGE.
Activity The activity was tested by binding Human ACE2 in functional ELISA assay (see figure below).
Purity >95%, as determined by SDS-PAGE and HPLC
Endotoxin Endotoxin content was assayed using a LAL gel clot method. Endotoxin level was found to be less than 0.1 ng/µg(1EU/µg).
Presentation Recombinant nCoV-2019 Spike S trimer protein was lyophilized from 0.2 µm filtered PBS pH7.4.
Reconstitution A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.
Storage The lyophilized protein is stable for at least 2 years from date of receipt at -20° C.
