Source:

Optimized DNA sequence encoding  Human Cystatin A- CSTA (ILE 2-PHE98) mature chain including a C-terminal His tag was expressed in HEK293 cells.

Molecular weight:

Recombinant Human Cystatin A (CSTA) is a protein consisting of 127 amino acid residue subunits, due to glycosylation migrates as an approximately as 17kDa band protein on reduced SDS-PAGE.

Purity:
>95%, as determined by SDS-PAGE and HPLC

Biological Activity:
The activity was tested by the ability to inhibit Cathepsin L cleavage of a fluorogenic substrate. The ED50 was measured to be 10nM

Endotoxin:
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/µg(1EU/µg).

Presentation:
Recombinant Cystatin A is lyophilized  from 0.2 μm filtered PBS solution,  pH7.2 ,  5% Trehalose.

Reconstitution:
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.

Storage:
Recombinant Human Cystatin A (CSTA) can be stored in working aliquots at 2° - 8° C for one month, or at -20°C to -70°C for twelve months.

Avoid repeated freeze/thaw cycles.

Usage:
This product is for research purposes only.It may not be used for therapeutics or diagnostic purposes.

Cystatin A

human cystatin A is an 11- kDa protein single chain intracellular cysteine protease inhibitor capable of inhibiting papain and cathepsins B, H, and L, as well as the cysteine protease activity of the major house dust mite allergen Der p1 . Cystatin A is expressed at very low levels in the liver ,cystatin A does not have a signal peptide for cellular secretion and is, therefore, retained within the cell instead of being secreted . Overexpression of cystatin A has been shown to result in reduced degradation of the cell cycle regulatory protein cyclin B in NIH/3T3 fibroblasts and inhibition of virus-induced apoptosis in a teleost cell line
