Molecular weight:

Recombinant human HIF-1α is a protein consisting of 260 amino acid residue subunits, migrates as an approximately 35kDa protein on reduced SDS-PAGE.

>95%, as determined by SDS-PAGE and HPLC
 
 
 
Endotoxin:
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/µg(1EU/µg).

 
Presentation:
Recombinant Human Hypoxia inducible factor 1 is lyophilized  from 0.2 μm filtered PBS solution,  pH7.2 ,  5% Trehalose.

              
Reconstitution:
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.

 
Storage:
Recombinant HIF-1α can be stored in working aliquots at 2° - 8° C for one month, or at -20°C to -70°C for twelve months.

Avoid repeated freeze/thaw cycles.

Hypoxia inducible factor 1

A key factor involved in the cell response to reduced oxygen levels is hypoxia- inducible transcription factor . HIF is a heterodimer that consists of a hypoxic response α subunit and the constitutively expressed subunit HIF-1β. In the presence of oxygen, HIF- α is bound to the von Hippel–Lindau tumor suppressor protein. This interaction causes HIF-α to become ubiquitylated and targeted to the proteasome, where it is degraded. Under hypoxic conditions, HIF-α becomes stable and binds HIF-1β to form the active HIF transcription factor, which binds to hypoxia- response elements activating the expression of numerous hypoxia-responsive genes. Stabilization of HIF-α in cancer cells is believed to be one of the most important mechanisms that promotes metastasis and, thereby, increases tumor aggressiveness .
