Optimized DNA sequence  encoding  extracellular domain of Mouse Proprotein convertase subtilisin/kexin type 9 (PCSK9) (GLN35 - GLN694) including an N-terminal His tag was expressed in HEK293 cells.


 
Molecular weight:

Recombinant Mouse PCSK9 is a protein consisting of 670 amino acid residue subunits,due to glycosylation migrates as an approximately 20kDa & 60kDa protein on reduced SDS-PAGE.


 
Purity:
>95%, as determined by SDS-PAGE and HPLC
 
Biological Activity:
The activity was tested by the ability of immobilized recombinant Mouse PCSK9  to bind recombinant human LDLR within a linear range of 10-100ng/ml in ELISA.
 
 
Endotoxin:
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/µg(1EU/µg).

 
Presentation:
Recombinant Mouse PSCK9 is lyophilized  from 0.2 μm filtered PBS solution,  pH7.2 ,  5% Trehalose.

              
Storage:
Recombinant  PCSK9 can be stored in working aliquots at 2° - 8° C for one month, or at -20°C to -70°C for twelve months.

Avoid repeated freeze/thaw cycles.

PCSK9

LDLR also binds proprotein convertase subtilisin/kexin type-9 (PCSK9), a secreted member of the mammalian proprotein convertase family of serine endoproteases . Bound PCSK9 disrupts the normal endocytic recycling itinerary of the LDLR and directs the receptor for degradation in endolysosomal compartments. PCSK9 binds in a Ca2+-dependent manner to the first of three Ca2+- binding EGF-like repeats (EGF-A) in the EGF precursor homology domain of LDLR and demonstrates increased binding affinity at acidic pH.
