CD26

First described in 1966, CD26/DPP4 is a serine protease found on the surface of diverse cells, with a catalytic activity that removes N-terminal dipeptides with proline or alanine at the penultimate position of various peptide substrates including inflammatory cytokines and chemokines . The proteolytic action of CD26/DPP4 is one of the multiple functions of the protein, with other functions including receptor activity, costimulatory functions, binding and interactions with various proteins, and a role in apoptosis. In line with its multiple functions, CD26/DPP4 is widely distributed in tissues, such as lung, spleen, liver, kidney, intestines, endothelial cells, bone marrow, and blood cells and is also present in serum as a soluble protein (sCD26/DPP4). Among blood cells, CD26/DPP4 is expressed predominantly, but not exclusively, on T lymphocytes, and most of the T cells expressing CD26/DPP4 belong to the CD4+ population. Additionally, the expression of CD26/DPP4 on T helper cells is tightly regulated upon activation, and T cell activation induces an increased expression of CD26/DPP4 for several days, which is more intense on Th1 cells than on Th2 cells . Taken together, CD26/DPP4 plays important roles in T cell functions, including activation, proliferation, migration, and memory response.
