Optimized DNA sequence encoding Human ENA-78 (CXCL5) mature chain was expressed in Escherichia Coli.

Molecular weight:

Human ENA-78, generated by the proteolytic removal of the signal peptide
and propeptide and has a calculated molecular mass of approximately 8 kDa.

Recombinant CXCL5 is a disulfide-linked monomeric protein consisting of 75 amino acid residue subunits and migrates as an approximately 8 kDa protein under non-reducing conditions and reducing conditions in SDS-PAGE.

Biological Activity:
Determined by its ability to chemoattract human neutrophils using a concentration range of 2.0-40.0 ng/ml.

Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/┬Ág(1EU/┬Ág).

Recombinant ENA-78 was lyophilized from a 0.2 ╬╝m filtered 20mM PB,100mM NaCl solution pH 7.5.

A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.

The lyophilized protein is stable for at least 2 years from date of receipt at -20┬░ C.
Upon reconstitution, this cytokine can be stored in working aliquots at 2┬░ - 8┬░ C for one month, or at -20┬░ C for six months, with a carrier protein without detectable loss of activity.

Avoid repeated freeze/thaw cycles.

Epithelial neutrophil- activating protein 78 (ENA-78) is a member of the CXC chemokines and acts as a potent chemoattractant and activator of neutrophil function. On stimulation in vitro, ENA-78 is highly expressed in many cell types. ENA-78 protein levels are strongly elevated in synovial fluid and blood of patients with rheumatoid arthritis. By in situ hybridization and immunofluorescence staining, ENA-78 has been recognized as a major CXC chemokine expressed in epithelial cells of the intestinal mucosa of patients with Crohnĺs disease, ulcerative colitis, and acute appendicitis.
