Molecular weight:

Recombinant CD10 is a monomer protein consisting of 718 amino acid residue subunits,  due to glycosylation migrates as an approximately 100 kDa protein on SDS-PAGE.


 
Purity:
>95%, as determined by SDS-PAGE and HPLC

 
Biological Activity:
Measured by its ability to cleave flourogenic peptide substrate Mca­-RPPGFSAFK(Dnp)-­OH. The specific activity was measured to be >1nMole/min/ug

 
Endotoxin:
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/µg(1EU/µg).

 
Presentation:
Recombinant CD10 is supplied as a 0.2 μm filtered solution in 20mM Tris,100mM NaCL, pH8.0 with 5% Trehalose

              
Reconstitution:
To prepare stock, reconstitute by gently mixing in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.DO NOT VORTEX

              
Storage:
Recombinant Neprilysin, as supplied, can be stored in working aliquots at 2° - 8° C for one month, or at -20° C for six months, with 0.1% HSA as carrier protein without detectable loss of activity.

Avoid repeated freeze/thaw cycles.

CD10

Neprilysin is a zinc metallopeptidase expressed at the cell surface of a variety of cells. The enzyme functions both as an endopeptidase with a thermolysin-like specificity and as a dipeptidylcarboxypeptidase. NEP has been shown to be involved in the degradation of enkephalins in the mammalian brain and the inactivation of circulating atrial natriuretic peptide. NEP has also been identified as the common acute lymphoblastic leukemia antigen (CALLA), and to be expressed on the surface of lymphocytes in some disease states. These and other observations have resulted in considerable clinical interest in NEP as a potential target for analgesics and antihypertensive drugs. NEP is also a major degrading enzyme of amyloid  peptide (A) in the brain, indicating that down- regulation of NEP activity, which could be caused by aging, can contribute to the development of Alzheimerĺs disease by promoting A accumulation.
