Optimized DNA sequence encoding Human Fibroblast Growth Factor-10 mature chain was expressed in Escherichia Coli.

 
Molecular weight:

Native human FGF10 is generated by the proteolytic removal of the signal peptide
and propeptide, this molecule has a calculated molecular mass of approximately 19 kDa.

Recombinant Fibroblast Growth Factor 10 is a disulfide-linked monomer protein consisting of 170 amino acid residue subunits, and migrates as an approximately 19 kDa protein under non-reducing and reducing conditions in SDS-PAGE.


 
Purity:
>96%, as determined by SDS-PAGE and HPLC

 
Biological Activity:
The ED(50) was determined by the dose-dependent  proliferation of
BaF3 cells expressing FGF receptors, and was found to be
<0.5ng/ml., corresponding to a specific activity of 2.0 x 106 Units/mg.

Endotoxin:
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/µg(1EU/µg).

 
Presentation:
Recombinant FGF10 was lyophilized from 0.2 μm filtered PBS solution, pH 7.0 .

 
Reconstitution:
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.

 
Storage:
The lyophilized protein is stable for at least 2 years from date of receipt at -20° C.
Upon reconstitution, this cytokine can be stored in working aliquots at 2° - 8° C for one month, or at -20° C for six months, with a carrier protein without detectable loss of activity.

Avoid repeated freeze/thaw cycles.

FGF10

FGF-10 is involved in the initial budding as well as the continuous outgrowth of vertebrate limbs, FGF10 mRNA is expressed preferentially in neurons but not in glial cells and may have a distinct role in the brain. human FGF-10 is mitogenic for fetal rat keratinizing epidermal cells but not for NIH 3T3 cells .Recombinant FGF10 induces the proliferation of human urothelial cells in vitro and induces the proliferation of transitional epithelium. FGF-10 is secreted by cultured mouse pre- adipocytes, prevention of FGF-10 signaling inhibits subsequent differentiation. The ability of embryonic fibroblasts derived from FGF-10 knock-out mice to differentiate into adipocytes is also impaired.
